Currently, a genetically edited form of herpes simplex virus to keep it out of the nervous system is knocking tests out of the ballpark according to studies from the University of Cincinnati and Northwestern University.
Researchers injected guinea pig subjects with the modified virus and results revealed a resulting reduction in genital lesions, less viral replication, and less viral shedding upon exposure to the live virus. The study revealed cross protection against HSV type 2 as well, which is relevant because a specific HSV-2 modification could be effective as well.
The researchers took out five codons from the viral genome with the hopes that the mutations would not reach the nervous system; the trial succeeded as the virus halted at the nerve terminal. The issue is if you continue to modify a virus, it can lose its ability to successfully replicate and provide a beneficial immune response.
These results prove particularly exciting because there have been 40 years of research into HSV-1 and HSV-2 and no vaccines have been approved. This is heavily attributed to herpesviruses possessing the ability to evade immune responses in the human body. I am interested to see if there are any unintended side effects, issues balancing the fine line between successful replication and avoiding the nervous system, and efficacy in humans.
