Can Rotavirus vaccine cause Intussusception in infants?

This case-control study looked into the risk of intussusception following Rotavirus Vaccination. Rotavac, a 3-dose oral rotavirus vaccine, was introduced as part of the universal immunization program in India in 2016. The investigators enrolled 970 infants with confirmed intussusception in an active hospital-based surveillance study at 27 hospitals in India. 589 of the infants were included in the analysis. The control cohort included infants who were admitted with illness unrelated to the gastrointestinal tract within 30 days before or after the admission of the matched case patient and were matched for age, sex and location. The study evaluated the relative incidence of intussusception among infants 28 to 265 days of age within three risk windows: 1 to 7 days, 8 to 21 days and 1 to 21 days after vaccination. Their results showed a relative incidence of 0.83 (95% CI, 0.00 to 3.00) for the first risk window and 0.35 (95% CI, 0.00 to 1.09) in the second risk window for the first dose. Similar results were also shown for the second dose. In conclusion, the data are consistent with no increase in intussusception risk amongst infants that received routine rotavirus vaccine in India proving no association between the vaccine and intussusception.
